Protein interaction causing rare but deadly vaccine-related clotting found

A mechanism that led some patients to experience cases of deadly clotting following some types of Covid-19 vaccination has been identified in new research.

In a recent paper published in Blood, scientists from the University of Birmingham funded by the National Institute for Health and Care Research and the British Heart Foundation have been able to identify how deadly blood clots, in the disease known as Vaccine-Induced Immune Thrombocytopenia and Thrombosis (VITT), occur.
Previous studies have shown that patients with VITT produce antibodies that stick to a protein called Platelet factor 4 (PF4) to create a large cluster of molecules called an immune complex. Following the development of a complex, platelets and cells of the immune system causing clotting and inflammation are activated, but the precise nature of what PF4 does in this event was unknown.

In this latest study the team used blood taken from healthy donors, as well as serum and plasma from patients with VITT, and have been able to learn for the first time how PF4 was directly involved in the activation of platelets and result in thrombotic events. By sticking to a receptor called c-Mpl on the surface of platelets, PF4 triggered the production of the small cells known to cause clotting.

Dr Richard Buka, Research Fellow in the Institute of Cardiovascular Sciences and co-lead author “As well as identifying a new way in which platelets are being activated in a potentially deadly manner in VITT, our research has also been able to find how this mechanism may lead to new drugs to protect against blood clots in VITT and blood clots in general.”

Variations on a drug used to treat bone marrow cancers could be developed to protect VITT patients from deadly clotting, the research also found.

The team used ruxolitinib, a drug used to treat some types of blood cancer, to block the receptor being triggered by PF4 following the vaccine-induced event. Although they note that the current form of the drug is unsuitable for use in VITT patients, the team nevertheless identified that blocking the pathway through ruxolitinib slowed down platelet aggregation and demonstrates a potential future way to protect patients from blood clots.

Mystery pneumonia tears through schools in China with 'many, many hospitalized' - in eerie echo of early days of COVID

A mystery pneumonia is reportedly tearing through schools in China. Hospitals in Beijing and 500 miles northeast in Liaoning are among those 'overwhelmed with sick children,' while school classes were 'on the verge of suspension', according to local news reports.

The children are presenting with unusual symptoms that include inflammation in the lungs and a high fever - but no cough or other symptoms usually associated with flu, RSV and other respiratory illnesses. The alert was issued late on Tuesday via ProMed - a large, publicly available global surveillance system that monitors infectious diseases.

It was a ProMed notification back in December 2019 that brought a mystery virus, later given the name of Covid, to the attention of many doctors and scientists, including officials at the World Health Organization.

WHO calls on China to reinstate masks, social distancing and staying home when ill amid mystery pneumonia outbreak

People in China should wear masks, socially distance and stay home if unwell amid an outbreak of a mystery respiratory illness, global health chiefs have warned.

Cases of 'undiagnosed pneumonia' have been detected at hospitals in Beijing and in Liaoning, a province 500 miles northeast of the capital.

Health facilities are 'overwhelmed with sick children' and classes are on the verge of being suspended according to local news reports.

Infected children are presenting with lung inflammation and a high fever, but not a cough or other symptoms that signal flu, RSV or another respiratory illnesses.

The situation prompted an alert from ProMed — a disease surveillance system which similarly sounded the alarm of a mystery infection in Wuhan in the closing days of 2019, which would later emerge as the global Covid pandemic.

The World Health Organization (WHO) has urged residents to don face coverings, stay away from ill people and remain at home if unwell. It has also called for the famously opaque nation to share 'detailed information' on the outbreak.

In one account on social media, which Newsweek was unable to independently verify, a video claimed Tianjin Children's Hospital was being overwhelmed with an average of 13,000 patients per day.

In Dalian, in China's northeastern province of Liaoning, children were receiving treatment in hospital lobbies due to the influx of cases, according to Taiwan-based FTV News on Tuesday. The report indicated that emergency rooms were overwhelmed and outpatient clinics no longer able to accommodate new patients.

The geographical spread of the cases, with affected areas in Beijing and Liaoning over 400 miles apart, suggests a wider outbreak than initially perceived.

According to Zhou Huixia, director of the children's medical center at the same hospital, the wave of infections began to intensify around China's National Day holiday in early October, according to the China Daily report. The outbreak has seen higher rates of antibacterial resistance and mixed infections compared to previous years, it said.

The World Health Organization (WHO) said it had seen data from China suggesting the spike in respiratory illnesses was due to common infections rebounding after the country's brutal lockdowns.

Chinese health officials say the outbreak - which is 'overwhelming' some hospitals and pushing schools to the brink of closure - is the result of a mix of infections caused by mycoplasma pneumoniae, RSV, adenovirus and the flu.

But the WHO is calling for full cooperation from China, which covered up the 2003 SARS outbreak and failed to alert the world to Covid for months, leaving countries flat-footed in their responses.

However, in a statement today, UKHSA chief executive Professor Dame Jenny Harries said despite China's official response, they are keeping an 'open mind'.

Doctors and health agencies in China believe Covid, RSV, flu and mycoplasma pneumoniae, a common bacterial disease also known as 'walking pneumonia,' are responsible for the surge.

They claim these illnesses are causing harsher illness because children's immunity was weakened during the country's strict lockdowns - which is not dissimilar to what happened in the US and UK last year.

But there are still doubts about China's transparency and many point to the eerie similarities between this outbreak and the initial weeks of the Covid crisis.

China covered up the original SARS epidemic in 2003 and its delay in reporting Covid in late 2019 left countries flat-footed in their responses.

DC area hospitals report uptick in sick visits as global concern for respiratory illnesses grows

The mysterious respiratory illnesses that are reportedly wreaking havoc in hospitals in China are now raising concerns for the medical community across the country and impacting Washington D.C., according to reports.

FOX 5 DC has learned that hospitals, emergency rooms, urgent care locations and clinics are reportedly seeing an uptick in sick visits.

The World Health Organization (WHO) is also reportedly reviewing data from China after video surfaced showing several groups of people apparently being treated for what’s being described as a mystery respiratory illness, predominantly impacting children.

On Thursday, Chinese health officials told the WHO that they have not detected any "unusual or novel diseases" regarding an increase in respiratory illnesses and pneumonia in children.

According to the WHO, outside scientists said the situation called for close monitoring, but they were not convinced that the recent spike in respiratory illnesses in China signaled the start of a new global outbreak.

The arrival of new flu strains or other viruses capable of triggering pandemics typically starts with undiagnosed clusters of respiratory illness, according to the WHO. Both SARS and COVID-19 were first reported as unusual types of pneumonia.
